Support development of monoclonal CHO cell lines expressing IgG antibodies. This work was dual focused, supporting both Pfizer proprietary CHO expression cell lines as well as technology development for multicopy antibody expression. My project comprised design/construction of plasmids, trasnfections, mAb culture expansion, AMBR evaluation and genomic stability studies.

Utilize structural proteomic approaches complemented with mutational analyses to map protein interactions between lipoprotein lipase (LPL) and its inhibitors angiopoietin-like protein 4 (ANGPTL4) and apolipoprotein CIII (APOC3). Aim to identify an inhibition-resistant LPL variant to translate to new gene therapy for patients with lipase deficiency. Aim to identify molecular mechanisms of LPL inhibition to develop novel intervention to treat heart disease due to accumulation of atherogenic triglycerides.

Aimed to determined the molecular mechanism by which a type 2 thioesterase, human TE2, modulates the product distribution of human fatty acid synthase (FASN) in cells. Optimized the expression and purification of several recombinant proteins, developed a protocol for the synthesis of acyl-ACP, as well as developed an enzyme assay to determine the TE2 activity against this biologically relevant acyl-ACP substrate. Identified several crystallization conditions for target proteins, optimized for crystal size and diffraction quality.Solved the native crystal structure of human TE2.
