Aimed to determined the molecular mechanism by which a type 2 thioesterase, human TE2, modulates the product distribution of human fatty acid synthase (FASN) in cells. Optimized the expression and purification of several recombinant proteins, developed a protocol for the synthesis of acyl-ACP, as well as developed an enzyme assay to determine the TE2 activity against this biologically relevant acyl-ACP substrate. Identified several crystallization conditions for target proteins, optimized for crystal size and diffraction quality.Solved the native crystal structure of human TE2.
