Helped optimize neuron tracing and puncta reconstruction protocols for general use in the lab. - Developed numerous scripts to assist with various analysis protocols, including over a dozen MATLAB scripts which interfaced directly with Imaris and drastically improved analysis efficiency, speed, and data throughput for the lab. - Trained and supervised multiple undergraduate research assistants on 3D reconstructions and other image analysis protocols.

- Used whole-cell patch-clamp electrophysiology in acute hippocampal slices to study the role of astrocytic glutamate in epileptiform activity. Showed pharmacologically that the “paroxysmal depolarizing shift” (PDS) induced by high extracellular potassium may be partially driven by swelling-induced glutamate release from astrocytes.- Developed lab protocols for confocal imaging of cell volume change in a hippocampal slice and subsequent image stack analysis protocols/macros in ImageJ, in order to investigate astrocyte volume changes in the hypoosmolar model of neuronal hyperexcitability. Demonstrated that, in contrast with published work, pyramidal neurons and passive astrocytes are similarly osmosensitive and both swell rapidly in response to a hypoosmolar insult, regardless of animal age, solution temperature or other factors.
